02

What is it?

Semaglutide is a long-acting analogue of the naturally occurring GLP-1 hormone. It is the active ingredient in several prescription-only medicines, each with its own authorised indication, strength and delivery form.

A product that uses the name semaglutide is not necessarily equivalent to Wegovy, Ozempic or Rybelsus.

03

Intended / recognised uses

Authorised / recognised uses

  • Specific UK-authorised semaglutide products are used for type 2 diabetes or weight management in eligible patients under their respective product licences.
  • The authorised indication depends on the brand and formulation; these products are not interchangeable without professional review.

Reported / other uses

  • Other uses discussed online may be off-label, unsupported or inappropriate. A clinical result from one authorised product cannot be assumed for an unverified presentation.
04

How it works

GLP-1 receptor activation increases glucose-dependent insulin secretion, suppresses glucagon when glucose is elevated, slows gastric emptying and influences appetite and satiety.

These actions can affect glucose control and body weight, but response varies.

05

Potential benefits / effects

  • Authorised semaglutide medicines can improve blood-glucose measures in type 2 diabetes and can support weight reduction or maintenance in appropriately selected patients.
  • The expected benefit depends on the licensed product, indication, baseline health and professional follow-up; outcomes are not guaranteed.

Potential benefit is not a guarantee of benefit for any individual.

06

Risks & warnings

  • Important risks include acute pancreatitis, gallbladder disease, dehydration and kidney deterioration related to vomiting or diarrhoea, and hypoglycaemia with insulin or sulfonylureas.
  • Rapid improvement in glucose can worsen diabetic retinopathy in some people. Sudden or worsening visual symptoms require prompt assessment.
07

Possible side effects

Common side effects

  • Nausea, diarrhoea, vomiting, constipation, abdominal pain and headache; reduced appetite and indigestion may also occur.
  • Low blood glucose is particularly relevant when used with insulin or sulfonylureas.

Serious or urgent concerns

  • Persistent severe abdominal pain may indicate pancreatitis.
  • Serious allergy, severe dehydration, gallbladder symptoms, significant hypoglycaemia or sudden visual change requires urgent medical advice.
08

Who should not use it / precautions

  • Pregnancy, planning pregnancy and breastfeeding require specialist advice and use should follow the authorised product information.
  • Previous pancreatitis, severe gastrointestinal disease, diabetic retinopathy, kidney or liver impairment and use in younger people require careful assessment.
  • Semaglutide should not be combined casually with other GLP-1 or weight-management medicines.
09

Interactions

  • Insulin and sulfonylureas increase hypoglycaemia risk.
  • Slower gastric emptying may affect absorption of oral medicines; clinically important oral drugs should be reviewed.
  • A pharmacist or prescriber should check all prescription medicines, over-the-counter products and supplements.
10
